Installation for: destructive analysis of leaf plant samples

Fresh samples (typically leaf material) are taken from plants cultivated in IPK-APPP and are rapidly frozen in liquid nitrogen. Up to 384 samples may be taken per day within a period of 2-3 hours; max. 768 samples may be harvested per project. Harvested deeply frozen plant material (typically leaves; samples of 100 mg - 4 g in 2 ml or 20 ml vials) are subjected to automated cryogenic (at – 60 °C) grinding to a fine powder and can be aliquoted into up to 6 (4) target vials (1.4 ml; feed range 10 – 100 mg +/- 10% tolerance). Input sample capacity: up to 96 vials; processing time: 28 hours. Processed (powdered) plant material is suitable for molecular or biochemical extractions (e.g. for DNA, RNA, protein/enzyme, metabolite analyses).Standard non-targeted metabolite profiling) is carried out using GC-MS. In brief, sample processing includes extraction of polar metabolites from 15 mg plant tissue, automated inline-derivatization of samples immediately before analysis and the measurement with an Agilent 7890 gas chromatograph coupled to a LECO Pegasus HT time-of-flight mass spectrometer. Results typically contain the relative quantity of app. 75 known metabolites and a similar number of unknown analytes.
